An impedimetric immunosensor for the selective detection of CD34+ T-cells in human serum

Chimeric antigen receptor T (CAR-T) cell therapy has led to a paradigm change in cancer treatment. To allow widespread use of this therapy, it is important reduce the costs associated with cells manufacturing whilst ensuring high-quality production. Online biosensors can help monitor real time effectiveness engineering CAR-T cells, possibility be integrated on workflow without need extract samples for off line analysis that require specialised personnel and expensive analytical equipment. In context, we present an innovative label-free impedimetric immunosensor detection CD34+ T-cells human serum. The sensor based screen-printed gold electrodes functionalised anti-CD34 antibody, via mixed self-assembled monolayer 11-mercaptoundecanoic acid 6-mercapto-1-hexanol, which recognizes CD34 expressed onto surfaces. exhibits high selectivity wide working range serum, both Faradaic non-Faradaic conditions, being respectively 230 – 1.4 × 103 mL−1 50 1 105 mL−1. This result, along performance validation batch flow-through operations, demonstrates applicability developed clinical use.
